Optimal Aspergillus fumigatus and Asp f 1 serum IgG cut-offs for the diagnosis of allergic bronchopulmonary aspergillosis

چکیده
The presence of IgG antibodies (Abs) to Aspergillus fumigatus (Af) is a crucial diagnostic criterion for allergic bronchopulmonary aspergillosis (ABPA). Although precipitation traditionally used document Abs, anti-Af serum levels can also be measured by enzyme immunoassay (EIA). However, there are insufficient data on the optimal cut-offs assess performance EIA method. This study aimed determine cut-off binding crude Af extracts or recombinant Asp f 1 (by ImmunoCAP®) and compare their efficacy ABPA diagnosis with Af-precipitating Abs. age distribution (Af-IgG) (Asp 1-IgG) was established using sera from 694 healthy controls (HC). Receiver operating characteristic analysis Af-IgG 1-IgG purpose performed in 306 Af-sensitized asthma patients (including 49 ABPA), were determined. An age-dependent decline observed HC. Thus, determined separately as 60 mg/L aged <55 years, 45 those ?55 years. For 1-IgG, 6.6 set regardless age. such testing allowed sufficiently good performance, Abs had better applicability ABPA. We EIA, which useful clinical settings where precipitating unavailable.

The 37-kDa recombinant protein Asp f 2, encoding an allergen of Aspergillus fumigatus, was expressed in a prokaryotic expression system and immunologically evaluated for its functional and structural properties. The open reading frame for a 310-amino-acid-long protein was shown to encode a signal peptide of 31 amino acids. A native 37-kDa culture filtrate protein and a 55-kDa mycelial glycoprot...

Allergic bronchopulmonary aspergillosis (ABPA) is an immunologically complex allergic disorder caused by the fungal pathogen Aspergillus fumigatus. Elevated levels of total immunoglobulin E (IgE), specific IgE, and IgG antibodies in sera are important immunodiagnostic criteria for ABPA. International reference standards or standardized immunodiagnostic assays are not available due to a lack of ...

Among the molds causing allergy, Aspergillus fumigatus (Af) constituted a major species present both indoors and outdoors. The antigens both secreted and bound to hyphae and conidia have been isolated for immunodiagnosis of allergic aspergillosis. The crude extracts have been used to demonstrate IgE antibody in vitro and for skin testing. The crude extracts contain many antigenic and non-antige...

Allergic bronchopulmonary aspergillosis (ABPA), which requires a set of criteria for diagnosis, occurs in atopic individuals, predominantly asthmatics. Oral corticosteroids are the cornerstone for the management of the disease. Allergic Aspergillus sinusitis (AAS), clinico-pathologically similar to ABPA, is also diagnosed with a set of criteria including demonstration of fungal elements in sinu...
